摘 要:目的通过大鼠孕期邻苯二甲酸二(2-乙基)己酯(DEHP)暴露,评价DEHP对子代大鼠发育的影响,初步探讨DEHP的发育神经毒性机制。方法成熟雌性Wistar大鼠从妊娠日起用10mg/(kg.d)、100mg/(kg.d)、500mg/(kg.d)及1000mg/(kg.d)DEHP连续灌胃染毒至孕19天。动态观察孕鼠一般自然情况及生产率、仔鼠的体重变化及仔鼠的行为畸胎学指标。结果1000mg/(kg.d)DEHP剂量组孕鼠食欲降低,体重增长受限,而其他各组孕鼠一般自然情况良好;与对照组相比,DEHP不同剂量组生产率受到一定影响,1000mg/(kg.d)DEHP剂量组孕鼠生产率为0,出现吸收胎的现象;10mg/(kg.d)、100mg/(kg.d)、500 mg/(kg.d)各剂量组仔鼠第1天、4天、7天、14天体重比对照组有降低趋势;第21天500 mg/(kg.d)DEHP组的仔鼠体重增长受限,差别具有显著性;仔鼠开眼时间、平面翻正、断崖回避、触须定位、听觉惊愕反射实验各剂量组仔鼠的达标时间均有稍落后于对照组的趋势;负趋地性反射实验显示500 mg/(kg.d)DEHP组与对照组相比,达标时间延迟,空中翻正实验100mg/(kg.d)、500 mg/(kg.d)组与对照组相比,达标时间延迟,差别具有显著性。结论2-乙基DEHP胚胎期暴露影响子代大鼠的发育,存在着一定的剂量反应关系。Objective Our purpose is evaluating the effects of developmental toxicity in offspring rats by embryonic exposure to Di(2-ethylhexyl-)phthalate(DEHP),and provide some basic data for the further study of developmental neurotoxicity induced by DEHP.Methods The mature female Wistar andro-rats were gavaged consecutively with the DEHP dose of 10mg/(kg·d),100mg/(kg·d),500mg/(kg·d) and 1000mg/(kg·d) from the first day until the 19th day of pregnancy.The general situation of pregnant rats and the birth rate were observed dynamically.After the offsprings were born,the body weight,teratology index in rat offsprings were observed.Results The pregnant rats in the 1000mg/(kg·d) DEHP-group showed low appetite and low weight growth,but there were no significant changes in the rats of the other groups.Comparing with the control,the birth rates were disturbed by the treatment of DEHP,1000mg/(kg·d) group is 0,and were observed the phenomenon of Absorbed fetal.On the 1st、4th、7th and 14th day after birth the body weight of rat offsprings in 10mg/(kg·d),100mg/kg/d,500 mg/(kg·d) groups were lower than the control,but there was no statistical significance.The weight growth on 21st birth day of offsprings in 500mg/(kg·d)group was limited.At the same time,teratology indexes were observed,and the time of eye opening,surface righting,cliff avoidance,positioning antennae,hearing shock reflection experiment in each group of offspring rates were slightly behind target standard time.Compared with the control,the 500 mg/(kg·d) groups showed a delayed time in negative geotaxis reflection experiment.The 100 mg/(kg·d) and 500 mg/(kg·d) groups in air righting reflex experiment showed a delayed time,too.Both the differences were statistically significant.Conclusions After embryonic exposure to Di(2-ethylhexyl)phthalate(DEHP) the develop-mental toxicity of rat offsprings appeared,and there is a certain dose-respons-e relationship.
