摘  要:目的:观察缺血性痴呆(VD)大鼠神经行为学及海马CREB的改变,并探讨两者间的关系。方法:采用双侧颈总动脉永久性结扎(2 vessels obstruction,2-VO)法建立血管性痴呆模型,8周后采用Morriss水迷宫实验测试大鼠记忆行为,海马HE染色检测神经元损伤,PCR及免疫组化检测海马CAl区CREB表达的变化。结果:模型组与正常组相比,大鼠认知功和神经元损伤程度明显,海马区CREB表达也明显降低(P<0.01)。结论:2-VO法成功制作了缺血性痴呆大鼠模型。模型大鼠脑内海马区CREB水平明显下降,其空间学习记忆障碍可能与海马区CREB表达下降有关。Objective: To observe the changes of neuroethology and contents of CREB in vascular dementia rat model,and investigate their relationship.Methods: Model group rats were performed permanent bilateral common carotid artery ligation(2-VO).Morris water maze and HE staining were used to detected the neuronal damage in CA1 zone of hippocampus,the contents of CREB were detected by RT-PCR and immunohistochemistry and analyzed with image analysis system after 8 weeks.Results: The cognitive function in model group was obviously impaired(P0.01).The contents of CREB in CAl zone was also decreased observably in model group(P0.01).Conclusion: With the 2-VO,we performed the VD model successfully.The expression of CREB in hippocampus of model group rats are decreased remarkably comparing to the normal group rats,which is contributed to the impairment of learning and memory in the rats of model group.
