摘 要:目的观察白术和黄芪的不同提取部位对小肠上皮细胞(IEC-6)增殖的影响。方法以系统溶剂分离法分别提取白术和黄芪的石油醚、乙酸乙酯、正丁醇、醇沉上清液和多糖部位;另将白术和黄芪以乙醇提取,D101大孔树脂吸附法得到40%和70%乙醇洗脱部位。MTT法检测白术和黄芪正丁醇部位、醇沉上清液、多糖部位、40%和70%醇洗脱液对IEC-6细胞增殖的作用。结果初步发现黄芪正丁醇部位、醇沉上清部位、糖复合物、40%和70%醇洗脱部位加药24h能促进细胞增殖;白术40%乙醇洗脱液部位加药24h能促进细胞增殖,白术正丁醇部位、糖复合物、40%和70%乙醇洗脱液部位加药48h能促进细胞增殖。结论白术和黄芪正丁醇部位、多糖部位、40%和70%醇洗脱液在加药后不同时间对IEC-6细胞有不同程度的促进增殖作用;采用系统溶剂分离法和大孔树脂吸附法提取白术和黄芪不同部位,可以用于细胞增殖实验,为进一步开展白术和黄芪在细胞水平上的药理研究提供参考。Objective To study effects of different extract parts from Rhizoma Atractylodis Macrocephalae (RAM)and Radix Astragali(RA)on proliferation of intestinal epithelial cells. Methods Polysaccharides, petroleum ether extracts, acetic ether extracts, N-butanol extracts and ethanol supernate from RAM and RA have gotten by means of systematic solvent extraction. Total saponins have obtained by alcohol (at the concentration of 40 % and 70 %) reflux extraction and purified by macroporous resin D-101. We observed the influence of N-butanol extracts, ethanol supernate, polysaccharides, and 40 % and 70 % ethanol eluants on the proliferation of intestinal epithelial cells by MTT assay. Results N-butanol extracts, ethanol supernate, polysaccharide, and 40 % and 70 % ethanol eluants from RA, and the sample from RAM eluted by 40 % ethanol promoted the proliferation of intestinal epithelial cells after treatment for 24 hours. After treatment for 48 hours, N-butanol extracts, polysaccharide, and 40 % and 70 % ethanol eluants from RAM promoted the proliferation of intestinal epithelial cells. Conclusion N-butanol extracts, ethanol supernate, polysaccharide, and 40 % and 70 % ethanol eluants from RAM and RA can be obtained by means of systematic sol- vent extraction and macroporous resin adsorption method, and they exert different degrees of effect on the prolifera- tion of intestinal epithelial cells, which will provide evidence for the research of pharmacological mechanism of RAM and RA.
