摘  要:目的研究青天葵组织培养中的污染问题,并提出防治措施。方法通过对污染的青天葵材料使用无菌水,生理盐水(0.9%NaCl),0.1%升汞,漂渍液和消毒片(次氯酸钠)清洗,对感染植物消毒效果进行研究,并考察了次氯酸钠的灭菌浓度及灭菌时间。结果青天葵组培物使用5%次氯酸钠清洗10min效果较好,可基本抑制污染,植物材料仍能正常生长;升汞灭菌虽可较彻底抑制污染,但对植物伤害较大,只用于对感染的球茎灭菌。结论通过污染控制研究,能提高青天葵组织培养的成功率。Objective To obtain effective approaches to solve the contamination problem during the process of tissue culturing of Nervilia fodii (Hance) Schltr. Methods Sterile distilled water (SDW), saline ( 0.9% NaCl ) , 0.1% mercuric chloride and different concentration of sodium hypochlorite were used as decontamination agents. The growth of the cultured materials after the treatments was recorded. Then the effect of sterile concentration and duration of sodium hypochlorite treatment were studied. Results It showed that 5% sodium hypochlorite with a duration of ten minutes was suitable for decontamination for the plant materials; 0.1%mercuric chloride also showed good effect, but the plants were killed as well. It showed good effect when treated the corms. Conclusion The study of decontamination could increase the survival rate of the tissue culture materials of Nerviliafodii (Hance) Schltr., which lays the foundation for setting up an in vitro propagation system ofNerviliafordii (Hance) Schltr..
