摘 要:目的通过检测VE对运动疲劳所致机体DNA损伤的保护效应,分析运动性氧应激与DNA损伤的关系,探讨预防DNA损伤的有效措施,为减缓运动性疲劳对机体的损害提供实验依据。方法选取24只昆明雄性小鼠,将其随机分为对照组(CG)、运动组(EG)、补充VE组(VEG)和运动+补充VE组(EVEG),每组6只。通过反复力竭递增负荷跑台运动建立小鼠运动疲劳模型,采用灌胃法给小鼠补充VE,用单细胞凝胶电泳(SCGE)技术检测小鼠外周血细胞DNA损伤,并测定血清中SOD、GSH活性和MDA含量。结果EG组DNA损伤程度显著高于CG、VEG和EVEG组,而EVEG与CG组DNA损伤无显著差异;EG组小鼠血清中SOD活性较CG和EVEG组有非常显著升高,EG组GSH和MDA含量显著高于其它3组。结论运动疲劳可致小鼠外周血细胞的DNA损伤,并使抗氧化酶系SOD、GSH和脂质过氧化产物MDA含量升高,VE对运动疲劳小鼠外周血细胞DNA氧化损伤的具有明显的保护效应。Objective In order to reduce the damage from exercise fatigue, we investigated the effect of VE on peripheral blood cell from DNA damage induced by exercise fatigue, and discussed the relationship of Exercise induced oxidative stress and DNA damage, including the solution keep away from DNA damage. Methods The mice were randomly divided into four groups , including CG,EG,VEG and EVEG. We built up the exercise fatigue model for mice through the protocol of repeated exhaustive treadmill running and use the single cell gel electrophoresis(SCGE) to detect the DNA damage of peripheral blood cell in different groups, meanwhile we measured the changes of SOD and MDA and GSH of the mice. The VE was taken through stomach using needle. Results It showed that the DNA damage of EG were obviously higher than the other three groups. The EVEG didn' t have significant difference compared with the CG. The activity of SOD in EG increased significantly compared with CG and EVEG, and its content of GSH and MDA were significant higher than the other three groups. Conclusions The exercise fatigue could result the DNA damage on peripheral blood cell, it also increase the value of SOD and MDA and GSH, VE has good effect to protect blood cell from DNA damage.
