摘  要:目的建立人血浆中长春西汀体内代谢物阿朴长春胺酸的LC-MS/MS分析测定方法。方法采用Diamonsil C18色谱柱(4.6mm×200mm,5μm);流动相为2mmol.L-1醋酸铵-甲醇(8:92);采用三重四极杆串联质谱以多反应离子检测(MRM)方式进行负离子检测。结果阿朴长春胺酸的定量下限为10.61μg.L-1,线性范围为10.61~424.40μg.L-1(r=0.9993,n=7),日内、日间精密度均小于6.8%,平均方法回收率为97.48%。结论所建方法灵敏、精密、准确,可用于阿朴长春胺酸的药动学研究。OBJECTIVE To develop a LC-MS/MS method for the determination of apovincaminic acid(AVA) , the primary me tabolite of vinpocetine(VP), in human plasma. METHODS The mobile phase was consisted of methanol-water including 2 mmol · L^-1 NH4Ac (8:92), Triple-quadrupole tandem mass spectrometer by muhiple reaction monitoring(MRM) mode via turbo ion spray ionization(ESI) was used. RESULTS The LLOQ was 10. 61μg· L^-1, and the linear calibration curve was obtained in the concentration range of 10. 61 -424.40μg· L^-1 ( r = 0. 999 3, n = 7 ). The intra and inter day variances were less than 6. 8%, and the average method recovery was 97.48%. CONCLUSION This method is sensitive, specific and rapid. Therefore, it can be used for pharmacokinetic study of apovincaminic acid.
