摘 要:目的:通过研究莫达非尼(modafinil,MOD)用药后小鼠各脑区谷氨酸含量的变化,探讨MOD中枢兴奋作用的可能机制。方法:MOD灌胃给药,测定给药后小鼠脑部各分区谷氨酸含量。结果:MOD使小鼠大脑皮层谷氨酸含量显著增加,该作用呈剂量依赖性,但对其他脑区谷氨酸水平无显著影响。结论:MOD用药后使小鼠大脑皮层谷氨酸含量明显升高,可能与其中枢兴奋作用机制有关。Objective:To determine the glutamic acid content in seven different brain regions of mice,and to explore possible mechanism of central neural system stimulation of MOD.Methods:After oral administration of MOD,the content of glutamic acid was assessed in mouse brain to discuss the possible central stimulation mechanism of MOD.Results:Glutamic acid content in mouse cortex significantly increased after treatment of MOD and had a significant correlation with the brain drug concentration.Conclusion:MOD is easy to get through the blood-brain barrier and wildly distributed in the mouse brain.MOD effectively increases glutamic acid level in mouse cortex,which can be probably involved in the central stimulation of MOD.
