摘 要:目的探讨肾性高血压大鼠(RHR)动脉收缩、舒张功能的改变及其对钾离子通道阻断剂反应的相应变化。方法20只SD大鼠随机均分为对照组及RHR组(n=10),RHR组用内径为0.2mm的银夹夹住大鼠左肾肾动脉起始部制作两肾一夹RHR模型,而对照组只分离左肾动脉不套银夹。以无创性套尾法于术前及术后5周每周测量一次血压、心率。RHR模型制成5周后经水合氯醛腹腔注射处死大鼠,分别取两组大鼠股动脉、腹主动脉行离体动脉环实验,观察两组大鼠股动脉、腹主动脉对血管活性物质KCl、去甲肾上腺素(NE)、硝酸甘油(NTG)以及钾离子通道阻断剂四乙铵(TEA)、4-氨基吡啶(4-AP)的反应变化。结果1周后,对照组、RHR组血压开始升高(P<0.05),3周时达平台期(P<0.01),但心率无明显变化。5周后与对照组比较,RHR组股动脉及腹主动脉对KCl、不同浓度NE的收缩反应明显增强(P<0.05),对TEA及4-AP的相对收缩反应明显减弱(P<0.05),对NTG(浓度为10-2mmol/L)的舒张反应明显减弱(P<0.05)。结论RHR动脉血管收缩反应增强,舒张反应减弱,对钾通道阻断剂TEA、4-AP收缩反应减弱,考虑RHR动脉功能变化可能与钾通道功能的变化相关。Objective To investigate the changes in arterial vasoconstriction and vasodilatation functions of renal hypertensive rats (RHR), and the effects of renal hypertension on the response to K+ channel blocker. Methods Twenty SD rats were randomized into two groups (10 each): RHR group and control group (left renal artery was isolated only). RHR model was established in RHR group with two-kidney, one-clip method (2K1C), with the diameter of the proximal and of the left renal artery reduced to 0.2mm internal diameter by a silver clip, while in control group, only the left kidney was isolated without clipping of the renal artery. Systolic pressure (SP) and heart rate were measured by standard tail-cuff technique before operation and after operation every week for 5 weeks. The animals were sacrificed by peritoneal injection of chloral hydrate 5 weeks after surgery. Femoral arteries and abdominal aortas were collected, and the effects of KCl, norepinephrine (NE), nitroglycerin (NTG), tetraethylammonium (TEA), and 4-aminopyridine (4-AP) on these arteries were observed with the isolated artery ring technique. Results One week after the operation, the SP elevated significantly compared with the control group (P0.05), and 3 weeks after the operation, the SP in RHR group reached the highest level (P0.01). No significant difference of heart rate was found between the two groups. Five weeks after the operation, the contractile response of femoral artery and abdominal aorta induced by KCl or NE increased significantly, while the relative contractile response induced by TEA (TEA/KCl) or 4-AP (4-AP/KCl) decreased significantly, and the vasodilatation response induced by NTG decreased significantly in RHR group compared with control group (P0.05). Conclusion The vasoconstriction response of arteries in RHR is increased, while the vasodilatation response is decreased. The vasoconstriction response induced by TEA and 4-AP is also decreased.
