摘 要:以羟丙基-β-环糊精(HP-β-CD)为手性流动相添加剂,提出了头孢羟氨苄对映体的高效液相色谱拆分方法。选用SinoChromODS-BP色谱柱(4.6mm×200mm,5μm),含16mmol.LHP-β-CD的甲醇-乙腈-水(10+10+80)混合溶液为流动相,pH为4.93,流速为0.30mL·min-1,紫外检测波长为232nm,进样量为10μL。试验结果表明:该法可使头孢羟氨苄的两对对映体得到良好的分离,分离度分别为1.51,1.53。Resolution of enantiomers of cefadroxil by HPLC was proposed by using the SinoChror ODS-BP chromatographic column (4. 6 mm×200 mm,5 μm)as stationary phase, and 16 mmol · L-1 hydroxypropyl -β- cyclodextrin (HP-β-CD) as a chiral agent added to the mobile phase which was a mixture of CH3 COH, CH3CN and H2O mixed in the ratio of 10+10+80. Acidity of the mobile phase was controlled at pH 4. 93 with a flow-rate of 0. 30 mL · min-1. UV detection at the wavelength of 232 nm was adopted for the determination, and volume of sample introducted was 10 μL. It was shown that the two pairs of enantiomers of cefadroxil were successfully separated by the proposed method and values of separation factor achieved were 1.51 and 1.53 respectively.
