摘  要:目的观察大气可吸入颗粒物一次气管滴注染毒后,大鼠血液炎症和内皮损伤指标的改变,探讨颗粒物所致心血管效应的生物标志。方法将32只体重为380~470 g的雄性Wistar大鼠随机分为4组,即空白对照组、颗粒物低、中、高剂量组(0,5,15和45 mg/kg);采用气管滴注染毒,24 h后处死动物,分类计数血细胞,测定血液炎性指标白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、超敏C-反应蛋白(hs-CRP)以及内皮损伤指标一氧化氮(NO)和血管性假血友病因子(vWf)水平。结果各染毒组大鼠血液白细胞总数明显升高,高剂量组达(7.94±1.15)×109/L、中性粒细胞百分比增加至(40.50±5.75)%;血清IL-6、hs-CRP水平与对照组比较明显升高(P〈0.05);中、高剂量组TNF-α水平较对照组明显升高(P〈0.05),各染毒组血清NO、vWf水平与对照组比较,差异无统计学意义。结论可吸入颗粒物染毒后可引起大鼠系统炎症反应,但未见内皮损伤指标的改变。Objective To observe the systemic inflammatory changes of blood,endothelial damages,and cardiovascular effects caused by PM10.Methods Thirty two male Wistar rats weighted 380-470g were randomly divided into one control group and three PM10 exposed groups.PM10were administered to the rats by intratracheal instillation at the doses of 5,15,45mg/kg·bw,respectively.The rats were sacrificed 24 hours after the treatment.The number of blood cells and the levels of IL-6,TNF-α,hypersensitive C-reactive protein(hs-CRP),nitric oxide and Von Willebrand factor in the serum were determined.Results The number of white blood cells,the percentage of neutrophils,the levels of IL-6,and hs-CRP in the serum increased significantly in all the PM10treated rats.TNF-α level in the serum increased markedly in 15 and 45mg/kg PM10 treated rats.No significant change was found in the levels of nitric oxide and Von Willebrand factor in the serum of PM10 treated rats.Conclusion The study showed that PM10 can cause systemic inflammation.However,there is no significant change in the blood indices for endothelial injury.
