摘 要:利用农杆菌LBA4404(pTOK233)转化水稻成熟胚愈伤组织,通过筛选获得抗性愈伤组织并再生出转化植株,转化频率为8.9%。转基因植株经分子生物学鉴定为阳性,外源基因gus在转化植株中表达亦提供了转化成功的证据。转基因植株的当代及T1代植株获得了种子。摸索了应用农杆菌转化水稻的方法、条件。利用干燥处理法抑菌并提高转化植株再生频率。Rice calli from mature embryos were transformed with Agrobacterium LBA4404(pTOK233). After selection and regeneration, transgenic plants were obtained. The frequency of transformation was 8.7%. PCR and Southern blot showed that the foreign genes had been transferred into plant chromosomes. X Gluc staining showed that foreign gene(gus) can express in plant cell. The seeds were obtained from transgenic plants and T 1 generation. Partial desiccation can inhibit Agrobacterium and increase the frequency of transformation. The methods were optimized, and the factors affecting transforming rice using Agrobacterium were studied.
