摘 要:目的观察非诺贝特对肥胖大鼠脂联素蛋白多聚体构成的影响。方法 SD大鼠随机分为普通对照组(NC组)、高脂对照组(HF组)和非诺贝特组(FF组)。喂饲高脂饮食制作肥胖大鼠模型,以非诺贝特30mg·kg^(-1)·d^(-1)灌胃4周,ELISA法测定血清总脂联素,非还原非加热变性Western blot检测血清、皮下及内脏脂肪组织中各聚合态脂联素蛋白表达水平,计算高聚体(HMW)脂联素比例。结果 HF组大鼠血总脂联素、血清和内脏脂肪中HMW脂联素及HMW比例、皮下脂肪中HMW脂联素低于NC组。FF组血清、内脏脂肪中HMW脂联素及内脏脂肪中HMW比例高于HF组(P<0.05)。结论非诺贝特能提高内脏脂肪和血清中HMW脂联素蛋白水平。Objective To investigate the effect of fenofibrate on adiponectin compostion in high-fatdiet (HFD) SD rats. Methods Rats were randomly divided into 3 groups (n= 15) : chow diet rats as NC group, HFD rats treated with vehicle and fenofibrate (30mg · kg-1 · d-1 ) as HF and FF group respectively. Total serum adiponectin were detected by ELISA. Adiponectin oligomer compostion were separated by Western blot under non-heat-denatured and non-reduced conditions. Results In HF group, total and high molecular weight (HMW) adiponectin, and the ratio of HMW to total adiponectin were all significantly reduced as compared with NC group (P〈 0. 05). Compared with HF group, fenofibrate showed a significant increase of serum HMW adiponectin, HMW adiponectin and the ratio of HMW to total adiponectin in visceral fat (P〈0. 05). Conclusions Fenofibrate could enhance HMW adiponectin level in visceral fat.
