摘 要:由于吐鲁番地区独特的气候特点,对吐鲁番果蝇和实验室野生型果蝇进行随机引物扩增DNA多态性的初步研究。结果表明:吐鲁番果蝇与野生型果蝇在DNA水平上的有差异性,并且优化出一种提取果蝇基因组DNA的方法。文章研究的目的在于探讨吐鲁番果蝇在DNA水平上的遗传多样性。The experimental materials used in the Tulu fan region in 2007 collected and cultured in the laboratory of genetics Tulu fan Drosophila period of time. The fruit fly Drosophila and the other different is that it can live in hot arid zone of. In order to explore the Drosophila Tulu fan level in the DNA genetic diversity, estimated Tulu fan Drosophila melanogaster with the wild--type level in the DNA of the difference. Through the extraction of live and frozen fruit fly Drosophila have genomic DNA, followed by agar gel electrophoresis. And select 20 kinds of 10 BP long oligonucleotide random primers, and then to PCR amplification of genomic DNA. The results showed that. the optimized extraction method to extract genomic DNA results very well, and live and frozen fruit fly Drosophila genomic DNA extraction is the same as the results. Of Turban and laboratory wild--type Drosophila melanogaster to DNA restriction fragment length polymorphism analysis (RLFP). Optimized method of PCR amplification can be a very good band effect, and Turpan Drosophila melanogaster with the wild--type, were brought in section 2 and 4 have a different take.
