摘　　要：目的:利用葡聚糖凝胶(Sephadex)LH-20层析柱料对橄榄苦苷粗提物进行精制纯化。方法:瑞典安玛西亚公司蛋白质快速层析系统(AKTA FPLC),层析柱为蛋白纯化仪所配(20mm×300mm),装柱物为葡聚糖凝胶Sephadex LH-20,流速1mL/m in,以50%乙醇为流动相,检测波长254nm;橄榄苦苷的含量检测采用HPLC法。结果:橄榄苦苷在进样量为2mL时,经过二次过柱,其纯度可达到82.9%。结论:葡聚糖凝胶LH-20可重复利用,再生简便,为指导橄榄苦苷的生产提供参考。Objective： To purify the oleuropein crude extracts by sephadex LH-20 column chromatograph.Methods：This experiment used fast protein chromatography system（AKTA FPLC）produced by Amersham,Sweden.The chromatography column（20mm×300mm）was matched with protein purification instrument.Sephadex LH-20 was used in the Fast Protein Liquid Chromatography columns.The mobile phase was 50% ethanol with a flow velocity of 1.0 mL per minute and the detection wavelength was 254nm.The content of oleuropein was determined by HPLC.Results：The purity of oleuropein was 82.9% after passing the column twice when the sample volume was 2 mL.Conclusion：Sephadex LH-20 can be re-used and the regeneration is convenient,it also provides a reference for the production of oleuropein.
