摘 要:目的:对肝动脉注射32P-玻璃微球(32P-GMS)的肝细胞损伤进行电镜形态计量,为肝癌的放射介入治疗提供形态学依据。方法:7只家猪经肝动脉注射直径为46~76μm的GMS,其中4只注射放射性32P-GMS,3只注射31P-GMS(对照)。术后2、4、16周分批取不同部位的肝组织进行常规电镜制样和电镜形态计量分析。结果:32P-GMS术后2周和4周时,核异常率及线粒体变异率均明显高于31P-GMS术后2周、4周及热球术后16周(P<0.001~0.01)。热球术后2周和4周时,线粒体体密度明显低于术后16周(P<0.01~0.02),但与冷球术后2周和4周无明显差异。线粒体的其它形态参数,各组间均无明显差异。结论:吸收剂量低于200Gy的32P-GMS,术后2周和4周时,肝细胞核及线粒体的损伤明显,随后逐渐恢复,至术后16周时恢复正常。说明肝细胞的损伤是短时而可逆的。Objective:EM morphometric study of hepacytes injury was carried out by hepatic arterial administration of phosphorus-32 glass microspheres( 32 PGMS) to offer the basis for interventional radiologic treatment. Methods:Seven domestic pigs were injected with 32 PGMS with a diameter of 46~76μm,through hepatic artery, in which 4 pigs were injected with radiating 32 PGMS,and the other 3 31 PGMS(contrast).The routine prepartion and EM morphometry on liver tissue from different parts was performed at week 2,4,and 16 after operation.Results:The ratio of nuclear abnormal and mitochondrial variation at week 2 and 4 after receiving hot spheres( 32 PGMS) was significantly higher than that at week 2 and 4 after receiving cold spheres( 31 PGMS) and week 16 after receiving hot spheres( P <0.01~0.001).The mitochondrial V v at week 2 and 4 after receiving hot sphere was obviously lower than that at week 16( P <0.01~0.02).No difference of mitochondrial V v was found between hot and cold spheres.There was no significant difference in regard to morphometric parameters of mitochondria.Conclusions:A serious injury of hepacytes nuclei and mitochondria was found at week 2 and 4 after operation and they recover gradually.If the absorbed does of 32 PGMS is less than 200Gy.The hepacytes at week 16 after operation recover to normal.It demonstrates that injury of hepacytes is reversible and lasted shortly.
