摘  要:目的:采用反相高效液相色谱法(RP-HPLC)测定大鼠血浆中替尼达普钠的含量,为其动物药代动力学研究提供方法学基础。方法:分析柱为4.0mm×200mm的HypersilBDS,C18,柱径10μm,流动相0.23‰KH2PO4∶乙腈=70∶30,含0.1%三乙胺,以H3PO4调节pH至7.6。流速1.0ml·min-1,柱温30℃,紫外检测波长为232nm,进样量10μl。结果和结论:血浆中内源性杂质不干扰替尼达普钠和内标物的测定,相对回收率大于94%,日内变异系数小于5%,日间变异系数小于10%,血浆中最低检测浓度为0.10mg·L1。该方法简便可靠。Objective∶A reverse phase-high performance liquid chromatographic(RP-HPLC) method for determination of tenidap sodium in plasma of rats is presented.Methods∶Sample was prepared for analysis by RP-HPLC.The analytical column was a hypersil BDS C 18 column (10μm,4.0mm×200mm),themobile phase was a 70∶30 mixture of 0.23‰ KH 2PO 4-acetonitrile containing 0.1% triethylamine and the pH was adjusted to 7.6,the flow rate was 1.0ml·min -1 and wave length for determination was 232nm.Results and Conclusion∶No interference by endogenous impurity was found.The linear range of the assay was 0.10~50.0mg·L 1 ,the measurable lowest limit of tenidap sodium was 0.10mg·L 1 ,and the recovery rate of the method was higher than 94%.The variation coefficience was less than 5% for within-day and less than 10% for between-day ,respectively.
