玫烟色棒束孢几丁质酶基因部分序列的克隆及分析

摘　　要：为了得到玫烟色棒束孢几丁质酶基因的保守区域,根据GenBank中几丁质酶基因的同源性序列设计简并引物,采用DNA和RNA分别作为模板扩增玫烟色棒束孢的几丁质酶保守区域。结果表明,DNA扩增的含有3个内含子,RNA扩增的不含内含子;所得到的氨基酸序列具有几丁质酶18族的典型特征,即1个酶作用活性位点和几丁质结合区。A pair of degenerate PCR primer was designed based on the conserved domain of chitinase of other fungi from GenBank, genomic DNA, and total RNA was used as template for the conserved regions of PCR, and the sequence was isolated from Isaria fumosorosea. Results showed that it contained three putative introns amplified from DNA, but no intron from RNA. The amino acid sequence had two typical characteristics of glycosyl hydrolase family 18, namely a chitin-binding site and a presumed active site of chitinase.
