摘 要:目的探讨解偶联蛋白2(UCP2)基因-866 G/A、A55V和3′非翻译区(UTR)插入/缺失(I/D)多态性与维吾尔族自然长寿的关联。方法以新疆和田地区191名维吾尔族自然长寿个体为研究对象,以53名75岁前自然死亡的同地区、同民族个体作为对照。采用PCR-限制长度片段多态性(PCR-RFLP)和PCR-直接测序法对UCP2基因-866 G/A、A55V和3′-UTR I/D多态性进行分型。结果启动子-866G/A变异AA基因型与长寿相关,A55V多态性和3′-UTR I/D多态性基因型、等位基因分布在长寿组和对照组之间无差异。结论 UCP2基因启动子-866G/A多态性与长寿相关。Objective To investigate the relationship between three polymorphisms within the Uncoupling protein -2(UCP2) gene ( promoter region - 866 G/A, A55V and 3 'untranslated region(3' - UTR I/D) and natural longevity in the Uygur population. Methods 191 healthy individuals over 90 years old and 53 control individuals who died in their 65 to 70 years were recruited. The polymorphisms within UCP2 gene, -866 G/A, A55V and 3'untranslated region I/D were genotyped by PCR- restriction fraction length polymorphism (PCR - RFLP) and PCR - Sequencing. Linkage disequilibrium and haplotype characteristics among the polymorphisms were analyzed. Results - 866 G/A variation was related to the longevity. There was no difference in the genotypes and alleles distribution of the polymorphisms of A55V and 3' - UTR I/D between longevity group and control group. Conclusion There was correlation between the polymorphism -866G/A of UCP2 gene and longevity.
