摘  要:目的研究大鼠发育期下丘GABAARβ2和γ2亚单位的表达规律。方法选用生后P1到P16天的正常大鼠,将下丘组织提取的GABAARβ2和γ2亚单位mRNA用RT-PCR半定量,原位杂交方法定位和定性。将引物与内参GAPDH一起进行PCR,产物的电泳结果在紫外分析系统下进行灰度分析比较。用非放射性寡核苷酸探针进行原位杂交。结果GABAARβ2和γ2亚单位在生后正常大鼠下丘内表达的总体趋势是非线性增加的,P7表达最低,P13为高峰,P16下降到与成年时的水平相当。差异有统计学意义(P<0.01)。下丘GABAARβ2和γ2亚单位的表达模式相似。结论GABAAR两种亚单位在生后大鼠下丘的表达是阶段性和波动性的。两种亚单位在下丘内同步表达。Objective To explore expressions of GABAAR β2 and γ2 subunits in the inferior colliculus of newborn rats. Methods mRNAs expressions of GABAAR β2 and γ2 subunits in the inferior coUiculus of newborn rats from 1 to 16 days were quantitated by half quantitation PT-PCR and located by in situ hybridization. The PCR were done with interior control GAPDH. The gray scales of the electrophoresis products of PCR were observed and analyzed under an ultraviolet analytical system. In situ hybridization was done by a non-radioactive oligonucleotide probe and the results were observed by a light microscope. Results Expressions of GABAAR β2 and γ2 subunits in the inferior colliculus were gradually increased in a nonlinear way. Expression level was the lowest at day 7, reached a peak at day 13 and dropped, to a level similar with that of adult rats at day 16. The differences were significant( P 〈0.01 ). Conclusion Expressions of GABA^R β2 and γ2 subunits in the inferior colliculus were undulatory and phasic in postnatal rats. The two subunits were co-expressed in the inferior colliculus.
