摘 要:目的:考察并比较葛根散等3首解酒方剂对乙醇致小鼠急性肝损伤的保护作用,为进一步研发与应用提供依据。方法:制备醉酒小鼠模型,采用各方剂大、中、小剂量灌胃干预,以蒸馏水为对照,检测实验动物TBA、ALT、AST、GSH、MDA、肝脏系数等指标,并比较分析各方对乙醇致急性肝损伤的保护效果。结果:与正常对照组比较,模型对照组小鼠各项指标有显著改变(P<0.01);与模型对照组比较,葛根散、葛花解酲汤、石膏汤能明显改善小鼠血清TBA、ALT、AST和小鼠肝组织GSH、MDA水平(P<0.05或P<0.01);与模型组比较,只有葛根散大剂量组小鼠肝脏系数明显降低(P<0.01);各项指标改善幅度最大的情况集中在葛花解酲汤或葛根散,其TBA、ALT、GSH等指标与石膏汤比较具有显著差异(P<0.01)。结论:酒精可导致小鼠细胞破坏和脏器肿胀明显,3首方剂对稳定肝细胞,减少肝细胞破坏具有重要作用,葛花解酲汤和葛根散对乙醇导致小鼠急性肝损伤的保护作用比石膏汤强。Objective: This study is conducted to examine and compare the protective effect of three Jiejiu recipes against acute liver injury induced by ethanol for the development and application in practice. Methods : After the establishment of mice drunkenness experiment model, mice were lavaged by three varying Jiejiu recipes of high-, medium-, and low-dosage, respectively, with distilled water treatment as control. The TBA, ALT, AST, GSH, MDA, liver index and other indexes are measured. The protective effect of three Jiejiu recipes against acute liver injury induced by ethanol are also compared. Results : All index in model control group mice were significant difference to that in control (P〈0.01). Contents of TBA, ALT and AST in serum and the levels of GSH and MDA from liver tissue were significantly amended compared with control (P〈0.01 or P〈0.01). The coefficients of liver from the Pueraria powder group treated with large- dosage were significantly lower (P〈0.01) than that in model group mice. Pueraria powder or Gehua Jiechen decoction had the most significant effect on TBA, ALT, GSH levels of experimential mice.Conclusion:Alcohol treatment can damage cell and organ swelling in experimential mice clearly. The above three recipes can reduce acute liver injury induced by ethanol in mice, the Pueraria powder and Gehua Jiechen decoction had some protective effects on hapatic injury, and their effects were superior to gypsum soup treatment.
