摘 要:目的探讨全反式维甲酸(all-trans retinoic acid,ATRA)对卵巢上皮性癌细胞株COC2干预后肿瘤细胞内糖代谢的变化及意义,进而从肿瘤能量代谢角度说明ATRA对卵巢上皮性癌的抑制及诱导分化作用产生的机制。方法以不同浓度及作用时间的ATRA对COC2细胞株进行干预,测定细胞质糖原、乳酸脱氢酶(lactate dehydrogenase,LDH)、琥珀酸脱氢酶(succinate dehydrogenase,SDH)的含量。结果COC2经5μmol/L及10μmol/L ATRA作用后,细胞内糖原含量及SDH的含量增加,LDH含量下降,并与时间呈正比,但两浓度间差异无统计学意义;经1μmol/L ATRA作用后与对照组比较无差异。结论5μmol/L及10μmol/L ATRA作用于COC2癌细胞株后,可产生细胞内糖原及SDH含量增加、LDH含量下降的改变,减缓了肿瘤细胞内的能量代谢,从而达到诱导肿瘤分化、抑制肿瘤生长的作用。Objective To investigate the effect of using all-trans retinoic acid(ATRA) on the change of intracellular glucose metabolism after in epithelial ovarian cancer in vitro so as to reveal the mechanism of ATRA anti-epithelial ovarian cancer from intracellular energy metabolism.Methods COC2 cell were treated with different ATRA concentrations at 1,5 and 10μmol/L in different time,and then intracellular glycogen,LDH and SDH were determined by biochemistry assays.Results The obviously increased intracellular glycogen and SDH,and decreased LDH level were observed after using ATRA 5μmol/L and 10μmol/L with time dependent manner,but there was no significant difference between ATRA 5μmol/L and 10μmol/L.Conclusion Our results suggest that the increased intracellular glycogen,SDH and decreased LDH level after using all-trans retinoic acid in COC2 cells in vitro may be related to a decrease in the intracellular anaerobic metabolism,an increase in the intracellular aerobic metabolism,and to induce cell differentiation and inhibit tumor cell proliferation.
