摘  要:目的探讨白细胞介素1受体拮抗剂(interleukin-1 receptor antagonist,IL-1Ra)基因可变串联重复序列(variable number of tandemrepeats,VNTR)多态性及白细胞介素1β(interleukin-1β,IL-1β)基因外显子5多态位点TaqⅠ与天津地区脓毒症的相关性。方法应用聚合酶链反应(PCR)法和限制性片段长度多态性(RFLP)方法 ,观察90例脓毒症患者、120例健康对照者的IL-1Ra基因VNTR基因多态性及基因IL-1β外显子5多态位点TaqⅠ的分布。结果正常对照组中A2型等位基因和A1/A2基因的分布频率与脓毒症组相比明显增加(8.3%vs 2.2%,P<0.05;16.7%vs 4.4%,P<0.05),IL-1β外显子5TaqⅠ多态位点等位基因频率及基因型分布与健康对照组差异均无统计学意义。结论 IL-1RaA2等位基因在脓毒症的发生发展过程中可能具有潜在的保护作用。Objective To investigate the association between interleukin-1 receptor antagonist(IL-1 Ra) gene VNTR polymorphism and interleukin-1β gene polymorphic site Taq I with exon S and sepsis in the population of Tianjin. Methods The VNTR polymorphism of IL-1Ra gene and polymorphic site of interleukin-1β gene with exon 5 were examined in 120 controls and 90 sepsis patients with polymerase chain reaction and restriction fragment length polymorphism methods. Results The frequency of the type A2 alleles and the A1/A2 genotype distribution in the normal control group were significantly increased compared with in the sepsis group(8.3% vs 2.2%, P 〈0.05; 16.7% vs 4.4%, P 〈0.05) ,for IL-1β gene,no significant difference was found on allele frequency and genotype distribution between two groups. Conclusion IL-1 Ra A2 allele may have the potentially protective effect on developing sepsis.
