摘 要:目的:观察中药不同配伍方式对糖尿病大鼠的影响,探讨影响七箭方药效的因素。方法:高脂饲料加小剂量链脲佐菌素注射法建立糖尿病大鼠模型,以改良的均匀设计法设计药物的5种不同组合方式,不同组合药物灌服30d,检测血糖、甘油三酯、胆固醇、血肌酐、尿肌酐、尿白蛋白,计算肌酐清除率,对实验结果进行回归分析。结果:试验组1-5比模型组血糖、甘油三酯、胆固醇、尿微量白蛋白明显降低,有统计学意义(P<0.05或P<0.01);试验组3-5比模型组肌酐清除率升高,有统计学意义(P<0.05或P<0.01)。根据回归模型分析对不同指标的影响因素,结果黄连小檗碱对血糖的影响最大;对甘油三酯的重要影响因素依次为天花粉多糖、黄连小檗碱、鬼箭羽黄酮;对胆固醇的重要影响因素依次为黄连小檗碱、天花粉多糖、鬼箭羽黄酮;对肌酐清除率的重要影响因素为三七皂苷;对尿微量白蛋白的重要影响因素依次为鬼箭羽黄酮、黄连小檗碱、三七皂苷、天花粉多糖。结论:改良的均匀设计法简便可行,对糖尿病大鼠有一定的改善作用,可为七箭方的优化提供实验依据。Object: To observe the influence of different combinations of Chinese medicine on rat diabetic index so as to optimize formula. Method: Diabetic rats were induced by high fat food and low dose of streptozotocin, modified uniform design was used to combine different effective fractions, five kinds of combinations were given by intragastric administration for 30 days, glucose, triglyceride, cholesterol, creatinine clearance rate, urinary albumin were detected, the results were analyzed by regression analysis. Results: Glucose, triglyceride, cholesterol, urinary albumin in the five experimental group reduced significantly(P0.05 or P0.01). Creatinine clearance rate in experimental group three,four,five increased significantly(P0.05 or P0.01). The Influential factors on different index were analyzed by regression model. The most important factors to aim at glucose was berberine. The important factors to triglyceride were radix trichosanthis polycose, berberine, Evonymus alata anthoxanthin. The important factors to creatinine clearance rate was Radix Notoginseng saponin. The important factors to urinary albumin were Evonymus alata anthoxanthin, berberine,radix trichosanthis polycose, Radix Notoginseng saponin. Conclusion: Modified uniform design could decrease experiment times and obtain best combination, exense and time were saved.
