摘 要:目的建立小鼠肠道外感染轮状病毒(Rotavirus,RV)动物模型,观察该模型动物发病时的细胞免疫功能状态。方法通过腹腔注射RV,建立小鼠肠道外感染RV的模型。分别在病毒接种后第3天及第7天通过摘除眼球取血,流式细胞仪检测T细胞亚群。同时处死动物,无菌取出肺脏、肝脏和脾脏进行组织病理学检查。设立正常对照组。结果小鼠肠道外感染RV成功(可见到肺脏、肝脏和脾脏非特异性病毒感染性炎性病变)。淋巴细胞亚群测定:实验组CD3、CD4及CD4/CD8比值均较对照组明显降低(P<0.01)。结论RV引起小鼠肠道外脏器肺脏、肝脏和脾脏组织病变的同时,机体免疫功能亦明显降低。Objective To explore the relationship between immunological mechanism and extra-intestinal disease of group A rotavirus(GARV) infection.Some BALB/c mice models infected by GARV had also been established so that to study extra-intestinal organs pathology and immunity function.Method GARV infected models were created in BALB/c mice by inoculating GARV into abdominal cavity.After 3 or 7 days,the pathological changes in extra-intestinal organs(lung,liver and spleen) were observed by light microscopy and subclass of T lymphocytes in peripheral blood were measured by Flow Cytometry respectively.Control group of ten mice was set up,too.Result In most mice inflammation changes were showed in lung,liver and spleen.CD3,CD4 and the ratio of CD4/CD8 were reduced obviously in study group compared with the control group(P〈0.05).Conclusion The decline of immune function may play an important role in GARV extra-intestinal infection.
