摘  要:目的:应用凝胶内差异显示电泳技术和质谱技术进行肝郁气滞型乳腺增生组织的蛋白质组研究。方法:提取正常乳腺组织及肝郁气滞型乳腺增生组织总蛋白,分别用Cy3或Cy5标记,每一对Cy3和Cy5标记样品都与一个Cy2标记的内标等量混合,上样于同一胶中进行电泳分离,经不同光激发下扫描得到不同样品的蛋白质组图谱。所获得的图谱经DeCydcr6.5软件进行分析,筛选表达量有差异的蛋白质进行质谱鉴定。结果:在肝郁气滞型乳腺增生组织中,有8个蛋白质表达水平具显著变化。其中S-腺苷谷胱甘肽水解酶、蛋白体5、超氧化物岐化酶、甘油醛-3-磷酸脱氢酶、过氧化物酶5、肽酰脯氨酰顺反式异构酶表达量增加,而三联组氨酸核酸结合蛋白1表达量显著下降。结论:8种差异蛋白质可能与肝郁气滞型乳腺增生疾病的发生与发展有关。Objective:Applying two-dimensional difference gel electrophoresis and mass spectrum to study the proteome of hyperplasia tissue of mammary glands due to liver qi stagnation.Methods:The two kinds of general protein were extracted from normal mammary glands and surgically resected hyperplasia tissue belonging to stagnation of liver qi,they were labeled with Cy3 or Cy5 respectively,each Cy3-labeled sample and Cy5-labeled sample were mixed with on the same 2-D gel along with a Cy2-labeled mixture of all samples as an internal standard.The proteome profile of different samples were got by scanning,which were analyzed by DeCyder 6.5 software.Results:The expression of 8 kinds of protein were obviously difference in hyperplasia tissue of mammary glands due to liver qi stagnation and those in normal mammary glands,the expression of S-formylglutathione hydrolase,proteasome 5,superoxide dismutase,glyceraldehyde-3-phosphate dehydrogenase,peroxiredoxin-5, peptidyl-prolyl cis-trans isomerase A increased,the expression of histidine triad nucleotide-binding protein 1 decreased.
