摘 要:目的:探讨护津方对细菌脂多糖诱导的急性肺损伤(ALI)大鼠肺组织金属基质蛋白酶2mRNA(MMP-2mRNA)及其抑制因子2mRNA(TIMP-2mRNA)表达的影响。方法:40只雄性Wistar大鼠随机分为4组:正常对照组、LPS模型组、地塞米松组和护津方组,每组再分为4小时和8小时两个亚组。分别用地塞米松和护津方在LPS诱导ALI模型前预处理大鼠。检测大鼠肺体指数,实时荧光定量PCR检测肺组织MMP-2mRNA、TIMP-2mRNA表达,并光镜观察肺组织病理形态变化。结果:与模型组相比,在两个时相点护津方组大鼠肺体指数显著降低(P<0.01或P<0.05)、MMP-2mRNA表达表达明显降低(P<0.01)及TIMP-2mRNA表达明显升高(P<0.01或P<0.05)。病理学观察,模型组大鼠肺组织出现大片出血及坏死,护津方组大鼠肺组织病理损伤程度较模型组减轻。结论:护津方对内毒素致急性肺损伤大鼠有保护作用,其机理可能与其能降低MMP-2mRNA的表达和升高TIMP-2mRNA的表达有关。Objective:To discuss the effects of hujin decoction on expression of metalloproteinase-2mRNA(MMP-2mRNA)and tissue inhibitor of metalloproteinase-2mRNA(TIMP-2mRNA)of lungs of rats with acute lung injury caused by LPS.Methods:40 rats were divided into 4 groups,normal,model,dexamethasone and hujin decoction groups,each group was divided into two subgroups after LPS-induced 4 and 8 hours,dexamethasone and hujin decoction groups were respectively given dexamethasone and hujin decoction before injecting LPS to induce acute lung injury.The lung's index was determined,the expression of MMP-2mRNA and TIMP-2mRNA of lungs were examined by real-time fluorescent PCR method,the histopathology changes of lungs were observed by light scope.Results:To compare with model group,hujin decoction could lowered the lung's index of 4 and 8 hours groups,the expression of MMP-2 mRNA of hujin decoction group was lowered,too,the expression of TIMP-2mRNA was raised obviously.There was large piece of pulmonary hemorrhage and necrosis was seen in the model group,hujin decoction could improve the pathological changes.
