摘 要:目的:确定乌头类中药中药效指标成分并测定其含量。方法:初步确定新乌头次碱可作为乌头类中药的药效指标成分。采用高效液相色谱法测定新乌头次碱的含量:色谱柱为AgilentZobaxXDBC1(8250mm×2.1mm,5μm),流动相为0.01%氨水溶液-乙腈(梯度洗脱),流速为1mL·min-1,检测波长为240nm。结果:新乌头次碱进样量在0.51~10.2μg范围内与峰面积积分值呈良好线性关系(r=0.9998);平均回收率为102.1%,RSD=2.2%(n=6)。结论:本方法简便、准确、重复性好,可用于乌头类中药的质量控制。OBJECTIVE: To establish the method for the content determination of medicinal components in Aconitum carmichaeli. METHODS: Benzoylmesaconine was preliminary considered as medicinal components of A. carmichaeli. HPLC was applied to determine the content of benzoylmesaconine and performed on Agilent Zobax XDB C18 column (250 mm×2.1 mm, 5 um). Mobile phase consisted of 0.01% ammonia-acetonitrile (gradient elution) with flow rate of 1 mL. min^-1 and the detection wavelength were set at 240 nm. RESULTS: The linear ranges of benzoylmesaconine was 0.51-10.2 ug. The average recovery of benzoylmesaconine was 102.1% (RSD=2.2%, n=6). CONCLUSION: The method is simple, accurate and reproducible for the quality control ofA. carmichaeli.
