黄柏、地榆、白及对溃疡性结肠炎大鼠组织中NF-κB和细胞因子表达的影响  被引量:50

摘  要:目的探讨中药地榆、黄柏、白及对溃疡性结肠炎(UC)大鼠肠组织中核因子NF-κB和血清中白介素1β(IL-1β)、白介素10(IL-10)表达的影响。方法将60只成年SD大鼠随机分为正常组、模型组、地榆组、黄柏组、白及组、巴柳氮组。除正常组外,各组均以三硝基苯磺酸(TNBS)/乙醇灌肠制作大鼠结肠炎模型。造模成功后,各组予相应处理,检测大鼠血清中IL-1β、IL-10含量,鼠结肠组织中NF-κBp65蛋白水平。结果地榆组、黄柏组、巴柳氮组大鼠血清中IL-1β水平均明显低于模型组,其中地榆组下调最明显。地榆组、黄柏组、巴柳氮组血清中IL-10水平与模型组相比明显升高,其中巴柳氮组升高最明显,地榆组、巴柳氮组效果更显著,而地榆组与巴柳氮组之间疗效相当。与模型组相比,各中药治疗组、巴柳氮组结肠组织中NF-κBp65蛋白水平均明显下调。治疗后,地榆组、巴柳氮组结肠组织中NF-κBp65蛋白表达水平较正常组明显下调,与巴柳氮组相比,地榆组下调肠组织NF-κBp65蛋白活性更显著。结论地榆、黄柏、巴柳氮均可显著降低IL-1β,升高IL-10水平,地榆、巴柳氮能明显下调NF-κBp65蛋白活性;黄柏对主要致炎因子和抑炎因子、NF-κBp65活性有一定的调节作用,对UC治疗可起辅助抗炎作用;白及对TNBS大鼠UC模型结肠组织中NF-κBp65蛋白表达有一定的调节作用,但对IL-1β、IL-10水平无明显调节作用。Objective: To investigate the effect of Cortex Philodendra, Radix Sanguisorbae, Rhizoma Bletillae on the expression of IL-1β, IL-10 and NF-κB in the rats with ulcerative colitis (UC) . Methods: Sixty male rats were randomly divided into normal control, model group, Cortex Philodendra treatment group, Radix Sanguisorbae treatment group, Rhizoma Bletillae treatment group and balsalazide group. Acute UC models were induced by TNBS in each group except the normal control group. The groups received relevant treatments. The levels of IL-1β, IL-10 were determined by ELISA, and nuclear factor (NF) -κB p65 activity was detected using Western blot. Results: Compared with model group, the level of IL-1β was significantly decreased in the Cortex Philodendra treatment group, Radix Sanguisorbae treatment group and balsalazide group, and Radix Sanguisorbae treatment group was the lowest among these groups; the level of IL-10 was significantly increased in these groups, the balsalazide group was the highest among these groups, but there was no significant difference between Radix Sanguisorbae treatment group and balsalazide group; the expression of NF-κB p65 was significantly reduced. The expression of NF-κB p65 in Radix Sanguisorbae and balsalazide treatment group was also lower than that in normal control group, and that in Radix Sanguisorbae treatment group was the lowest. Conclusion: Cortex Philodendra and Balsalazide can inhibit the expression of NF-κB p65, IL-1β and promote the expression of IL-10. Radix Sanguisorbae can increase the level of IL-10, and decrease the level of IL-1β, but can not inhibit the expression of NF-κB p65. Rhizoma Bletillae can inhibitthe expression of NF-κB p65.
