摘 要:目的探索自体骨髓间充质干细胞(BMMSCs)不同移植途径对脑冷冻伤大鼠神经功能改善的影响。方法将Wistar大鼠自体骨髓间充质干细胞在体外扩增并经Brdu标记后,通过颈内动脉和脑室系统注射移植入冷冻伤脑水肿动物体内,以免疫组化和神经功能评分评价BMMSCs自体移植的疗效。结果经颈内动脉和脑室系统注射的Brdu标记的大鼠BMMSCs细胞可向脑损伤区域迁移。经颈动脉注射BMMSCs治疗组和经脑室注射BMMSCs治疗组Brdu阳性细胞的数量和神经功能恢复的程度均高于常规治疗组,且有明显统计学差异(P<0.05)。两移植组BMMSCsBrdu阳性细胞的数量和神经功能恢复的程度相比无明显差异(P>0.05)。结论本实验研究提示,BMMSCs通过循环系统和脑室系统移植后可向脑损伤区迁移而发挥治疗作用,但二者无统计学差异。Objective To explore the effect of autogenous bone marrow mesenchymal stem cells (BMMSCs) transplanted with different ways on improvement of nervous function in the rats with freeze-induced brain edema. Methods After BMMSCs derived from the Wistar rats were proliferated by culture, BMMCS were marked with Brdu in vitro. The Brdu-marked autogenous BMMSCs were injected into the internal carotid artery or the ventricular system in the rats with freeze-induced brain edema. The nervous function was assessed by neurological score and the brain tissues were observed by immnohistochemical staining in all the rats. Results The Brdu-marked BMMSCs derived from the rats immigrated into the injured brain areas after the injection into the internal carotid artery or the ventricular system. The number of Brdu-positive cells in the injured brain areas and the neurological scores in both the BMMSCs transplatation groups were significantly more than that in the control group (P0.05). There was insignificant difference in the number of Brdu-positive cells and the neurological scores between both the BMMSCs transplantation groups (P0.05). Conclusion The autogenous BMMSCs marked with Brdu can immigrate to the injured brain area and exert the therapeutic effect on the injured nervous function after the injection of them into internal carotid artery and the cerebrally ventricular system in the rats with freeze-induced brain edema.
