摘  要:目的探讨翼点入路在构建视神经损伤动物模型实验中的可行性。方法成年健康猫36只,按损伤时间分为正常、6h、1d、3d、7d、14d6个组,每组6只。模仿临床上翼点入路显微手术,暴露视神经,以无创血管夹夹持20s,于此后不同时间取视神经标本,应用免疫组化染色检测视神经胶质纤维酸性蛋白(GFAP)、神经丝蛋白(NF)及髓鞘碱性蛋白(MBP)表达,观察视神经损伤后的病理改变。结果视神经损伤后损伤侧瞳孔较正常瞳孔变大,直接对光反射消失,间接对光反射存在,直接钳夹损伤区GFAP、NF和MBP染色均减弱。结论采用翼点入路构建视神经损伤动物模型有临床及理论依据。视神经损伤后直接损伤区星形胶质细胞死亡,NF减少,髓鞘脱失。Objective To investigate the feasibility of establishing optic nerve injury animal model by the microsurgery through pterional approach. Methods Thirty cats were averagely divided into 6 groups of 6 animals each, i.e. posttraumatic 6 hours, and 1, 3, 7 and 14 days groups and normal control group. The optic nerve near the optic forman was squeezed for 20 seconds in each cat except the animals of the normal control group after a side of the optic nerve was exposed by microsurgery which simulated the clinical microsurgery trough pterional approach. The pathological changes in the optic nerves were observed by immunohistochemistry staining in order to determine the expressions of glial fibrillary acidic protein (GFAP), neurofilament protein (NF) and myelin basic protein (MBP). Results The pupils ipsilateral to the injured optic nerve, in which the direct light reflex disappeared, and the indirect light reflex existed, were dilated. The expression of GFAP NF and MBP reduced in the injured optic nerves. Conclusions The model of the optic nerve injury may be established by the microsurgery trough pterional approach in cats. The astrocytes and NF reduce, and the demylination occurs in the optic nerve injured by the microsurgery.
