牛膝总皂苷的制备及其保护尿酸钠致血管内皮细胞损伤的作用

摘　　要：目的:评价牛膝总皂苷抗急性痛风活性,为进一步深入研究牛膝皂苷的药效作用奠定基础;方法:建立香兰素-冰醋酸-高氯酸比色法测定牛膝总皂苷的含量;采用溶剂法制备总皂苷提取物;用尿酸钠(MSU)体外直接刺激血管内皮细胞(HUVEV)损伤的急性痛风模型,MTT法评价牛膝皂苷保护HUVEC活力的作用。结果:牛膝皂苷含量测定方法简单、灵敏准确,牛膝中总皂苷的含量为1.74%,总皂苷提取物纯度为70.9%,转移率为70.7%,对MSU致HUVEC的损伤具有显著的保护作用。结论:溶剂法制备牛膝皂苷工艺简便,纯度高;牛膝总皂苷具有抗急性痛风作用。Objective ： Evaluate the anti - acute gout effects of the saponins of Achyranthis bidentatae, to further study its mechanism for the role of anti - acute gout effects. Methods ：The content of saponins was determined by UV spectrophotometric method with Vanillin - Acetic acid and perchloric reagen, Solvent method was used to extract total saponins in Achyranthis bidentatae, MTF method was used to analyze the effects of the saponins on inflammatory functions HUVEC induced by MSU. Results： The content determination method is simple, the content of the ponins in Achyranthes bidentata was 1.74% and the total saponin extract purity was 70. 9% , and the total saponins showed significantly protective effects on HUVEV stimulated by MSU. Conclusion： The process of extacting total saponins by solvent method was simple, the saponins in Achyranthes bidentata showed the effect of the anti -acute gout.
