摘 要:[目的]建立一种适合玉米成熟期茎秆木质化、纤维化程度高,材料硬度大等结构特点的石蜡制片方法。[方法]以成熟期玉米基部茎秆节间的部分组织为材料,在传统石蜡切片方法的基础上,结合玉米成熟期茎秆的结构特点,对脱水、透明、透蜡、包埋、染色等重要制片环节进行了一系列的研究,得到了适合玉米成熟期茎秆的石蜡制片方法。[结果]玉米茎秆石蜡制片方法较传统方法整体上缩短了制片周期,结合玉米成熟期茎秆水分含量低减少脱水时间、采用蜡屑逐级透入弥补了传统透蜡不能使皮层致密的厚壁细胞组成的机械组织透蜡完全的缺点,并根据玉米成熟期茎秆木质化程度高的结构特点缩短染色周期,获得了厚度为10μm且染色清晰、组织结构完整的玉米成熟期茎秆横切切片。[结论]该方法进一步从解剖学上观察和研究玉米的生长机制和品种鉴定提供基础,同时为成熟期单子叶禾本科植物茎秆的石蜡切片技术提供借鉴。[Objective] The method of paraffin section suitable for corn stalk in a maturity stage with structural characteristics such as high degree of lignification and fibrosis,high hardness materials,was established.[Method] The tissue between two internodes at basal part of maturity corn being taken as experimental material,s of the suitable method of paraffin section of corn stalk was formed through a series of the improvements in main linkage such as dehydration,transparent,transparent wax,embedding,staining,etc,based on the traditional method and combined with structural characteristics of corn stalk in maturity stage.[Results] The improved method shortened the production cycle as a whole compared with the traditional method;the time in linkage of dehydration was reduced because of the material containing low content of moisture.The application of the measure wax debris penetrating into material level by level,instead of traditional one,corn stalk paraffin method than of combined with corn stalk maturity to reduce dehydration time of the wax level-up the penetration can make thick-walled cells completely being filled in with wax.The dyeing cycle of material was shortened because of its structural characteristics of a high degree of lignification in the maturity stage.Thus,10μm cross-section slice of maturity corn stalk,with clearly staining and complete organizational structure,was prepared.[Conclusion] The basis of further anatomical observation and study on corn growth mechanism and the identification of variety was provided,while the technical reference for the paraffin section of the cereal plant stalk in maturity stage of monocotyledon was supplied.
