摘  要:[目的]研究一种新整体透明与制片技术。[方法]将酒精溶液保存的千金藤幼嫩种子和高粱新鲜的雌蕊与珠心分离出来后,用84消毒液溶液对其进行浸泡、透明,然后按照3种制片方法进行制片处理和封片。[结果]在普通显微镜下不仅能观察到千金藤幼嫩种子中不同发育时期的球形胚和心形胚,还能观察到高粱雌蕊的特征、珠心表面的珠心突、胚囊和承珠盘;在荧光显微镜下可观察到高粱珠心内的胚囊和承珠盘的荧光图像。[结论]使用84消毒液作为整体透明技术的透明剂,不仅能观察保存后颜色变深或新鲜的植物材料,还能简化制片过程、缩短制片时间。[Objective] The research aimed to study a new whole clearing and preparation technology.[Method] After younger seeds of Stephania sp.,preserved in the alcohol solution,as well as the fresh pistil and nucellus of Sorghum bicolor were separated,they were soaked and cleared by the solution of 84 disinfectant.And then the plant materials could be mounted by using three methods.[Result] The globular embryos in different stages and heart-shaped embryos of Stephania sp.as well as the pistil,nucellus,embryo sac,hypostase of Sorghum bicolor could be observed under a microscope.And the embryo sac and hypostase of Sorghum bicolor could be observed under a fluorescence microscope.[Conclusion] As the disinfectant of whole clearing technology,84 disinfectant could not only observe the dark preserved or fresh plant materials,but it also could simplify the preparation process and shorten the preparation time.
