摘 要:[目的]探讨提取云南松毛虫蛹甲壳素的试验条件,为相关产品的开发提供参考依据。[方法]采用单因素试验法和正交试验法研究云南松毛虫蛹壳中甲壳素的提取技术,对试验结果进行了相关性分析、方差分析和q检验,并对不同条件下的N含量和甲壳素产率做线性拟合。[结果]云南松毛虫蛹壳甲壳素提取的最佳提取工艺为:1)脱有机物质(蛋白质):NaOH质量分数为8%,浸泡温度为90℃,浸泡时间为10h;2)脱矿物质:HCl质量分数为2.5%,浸泡温度为40℃,浸泡时间为22h;3)脱色:H2O2质量分数为11%,浸泡温度为80℃,浸泡时间为2.5h。[结论]制得的甲壳素为白色片状固体,N含量为6.23%、灰分含量为0.22%、水分含量为6.28%、产率为29.53%,产品质量指标均达到食品级甲壳素的标准。[Objective] To explore the extraction conditions of chitin from the pupae of Dendrolimus houi,and provide a reference for the development of corresponding products.[Method] The extraction technology of chitin from the pupae of Dendrolimus houi was studied using single-factor test and orthogonal experimental design,and then the correlation analysis,variance analysis and q test were conducted.The nitrogen content and the production ratio of chitin at different conditions were linear fitted.[Result] The optimum extraction conditions were as follows:1) removal of organic substances:8% NaOH at 90 ℃ for 10 h;2) demineralize:2.5% HCl at 40 ℃ for 22 h;3) decolorize:11% H2O2 at 80 ℃ for 2.5 h.[Conclusion] The obtained chitin was white solid sheet with 6.23% nitrogen,0.22% ash and 6.28% H2O,and the production ratio was up to 29.53%.The product quality is up to the criterion of food grade.
