摘  要:本实验通过改变体外受精精子浓度(0.5×10^6、2×10^6、8×10^6个/mL),精卵共培养时间(12h、18h、24h),体外受精96h后是否更换培养液(IVC)以及氧气的含量(5%CO2和5%CO2、5%O2、90%N2)等培养条件,观察牦牛精子对黄牛卵母细胞受精后卵裂和早期胚胎发育状况.结果显示精子浓度为2×10^6个/mL、精卵共培养24~26h、培养96h后要更换培养液,在5%CO2、5%O2、90%N2的条件下培养,受精和早期胚胎的发育.The objective of this study is to determine the effect of in vitro fertilization (IVF) conditions on cleavage rate and early development of embryos derived from bovine oocytes fertilized by yak sperm. These conditions include sperm concentrations (0.5×10^6, 2×10^6, 8× 10^6 sperm/mL) in IVF medium, sperm-egg co-incubation time (12 h, 18 h, 24 h) and replacement of culture medium at 96 h post-insemination (hpi), oxygen tension (5% CO2 and 5% CO2, 5% O2, 90% N2). The results show that there is a significant increase on the cleavage rate and blastocyst rate when sperm condition is 2×10^6 sperm/mL in IVF medium, sperm-egg co-incubation time for 24 h, culture medium replaces at 96 hpi and the embryos are cultured in 5% CO2, 5% O2, 90% N2.
