摘  要:目的:从桑叶中分离纯化获得均一多糖PMP12,并研究其组成及初步结构。方法:桑叶经热水提取乙醇分级沉淀,脱蛋白、脱色,经DEAE-纤维素和SephadexG-100凝胶柱层析,得到一个均一多糖组分PMP12,采用气相(GC)、高效液相(HPLC)、红外(IR)、核磁(NMR)、Smith降解和糖醛酸还原等方法分析其组成和初步结构。结果:PMP12由鼠李糖(Rha)、阿拉伯糖(Ara)、半乳糖(Gal)和葡萄糖醛酸(GluA)组成,其摩尔比为Rha:Ara:Gal:GluA=1:1.56:1.57:1.08;PMP12主链主要是以β-1→3糖苷键连接的鼠李糖,侧链主要是以β-1→2糖苷键及β-1→4糖苷键连接的阿拉伯糖、半乳糖和葡萄糖醛酸。结论:测定分析了桑叶多糖PMP12的组成和初步结构,为桑叶多糖的进一步研究提供依据。Objective:To study the isolation,the composition and characterization of polysaccharide PMP12 from Mulberry leaves.Methods:Mulberry leaves were extracted by boiling water.The polysaccharide in the filtrate was precipitated fractionally by alcohol.The protein in the product was removed by TCA and it was further purified by DEAE ion exchange cellulose (DEAE-52)and SephadexG-100,received PMP12.The composition and characterization of Mulberry leaves polysaccharide were researched by GC,HPLC,IR,1H-NMR,Smith degradation and uronic acid reduction and so on.Results:PMP12 is made up of rhammose,arabinose,galactose and glycuronic acid with the molarity rate of 1 ∶1.56 ∶1.57 ∶1.08;In the PMP12,the main chain is made up of β-1→3-linked-rhammose.The side chains are β-1→2 and β-1→4-linked-arabinose,galactose and glycuronic acid.Conclusion:The structure of PMP12 was first determined from mulberry leaves.These work could establish foundation for further study of Mulberry leaves polysaccharides.
