摘 要:[目的]观察1年生黄芪超微粉对环磷酰胺所致小鼠免疫抑制和肾脏毒副作用的影响.[方法]取昆明种小鼠随机分为正常对照组、模型组及小、大剂量1年生黄芪超微粉组,分别灌胃给予自来水或2,4 g/kg 1年生黄芪超微粉混悬液,连续11 d,第5,10日腹腔注射给予正常对照组等量生理盐水,其余各组均腹腔注射给予环磷酰胺80 mg/kg,第11日测定每组小鼠免疫器官指数、外周血白细胞计数和血尿素氮含量;取昆明种小鼠按照上述方法分组并给药,第3,6,9日除正常对照组外各组均腹腔注射给予环磷酰胺80 mg/kg,第10日测定肾组织丙二醛含量.[结果]与正常对照组比较,模型组小鼠胸腺、脾脏指数及外周血白细胞计数均明显降低,肾组织丙二醛含量明显升高;与模型组比较,小、大剂量1年生黄芪超微粉组胸腺指数及外周血白细胞计数均明显升高,肾组织丙二醛含量明显降低,大剂量1年生黄芪超微粉组血尿素氮含量明显降低.[结论]1年生黄芪超微粉可提高环磷酰胺所致免疫抑制小鼠免疫功能,减轻环磷酰胺对肾脏的毒副作用.OBJECTIVE To observe the effects of micro-milling of the annual Astragalus(MMAA) for the immunosuppression and nephrotoxicity induced by cyclophosphamide(CTX) in mice.METHODSThe Kunming mice were randomly divided into the groups of normal control,model,small and large dosage of MMAA.The mice in small and large dosage groups of MMAA were performed by intragastric administration of 2,4g/kg of MMAA for 11days,and in normal control and model groups were administrated with the same volume water.At the 5th and 10th day,the mice in all groups except control group were administrated by intraperitoneal injection with 80mg/kg of CTX,the same volume of physiological saline in control group.At the 11th day,the weights of immune organs,white blood cell counts(WBC) and the blood urea nitrogen(BUN) were detected.Kunming mice were selected and processed by the same methods of above-mentioned.On the 3rd,6th and 9th day,the mice in all groups except control group were intraperitoneally injected wiht CTX 80mg/kg,same volume physiological saline in control group.On the 10th day,the MDA levels in renal tissue was detected.RESULTSCompared with normal control group,in immunosuppressive mice,the weight of thymus and spleen,the WBC were decreased significantly.Compared with model group,the thymus weights and the WBC of mice in small and large dosage groups of MMAA were increased and the renal tissue MDA content markedly decreased,and the blood urea nitrogen content were significantly decreased in large dosage group of MMAA.CONCLUSIONMMAA has the effects of improving immune and antagonizing renal toxicity caused by cyclophosphamide in mice.
