摘 要:为了了解实施湖羊保种措施后群体遗传多样性状况,试验根据绵羊6号染色体遗传图谱和多胎FecB基因有关资料,选择位于绵羊FecB基因附近的6个微卫星位点(BMS2508、LSCV043、300U、GC101、Bulge5、471U)作为标记,分析其在湖羊保种群和普通商品群以及带有湖羊血统的中国美利奴肉用多胎品系杂交群中的遗传多样性。结果表明:6个微卫星位点均为多态性位点,在3个绵羊群体中分别检出34个、33个和29个等位基因,其中21个等位基因为3个群体所共有;平均多态信息含量分别为0.5054,0.4559,0.5329;保种群的有效等位基因数、遗传多样性和杂合度等均高于普通群,表明湖羊保种群具有较高的基因多样性。结果说明湖羊保种效果显著,但同时应加强稀有频率基因的保护。To study the genetic diversity and analyze the effect of conservation species, six microsatellite markers which on 6th chromosome were selected according to the genetic map and prolificacy molecular marker named FecB gene of sheep. These microsatellite markers are LSCV043, BMS2508, GC101, 300U, Bulge5 and 471U, respectively. And they were closely linked to sheep FecB gene.In this report, sheep DNA samples detected from Hu sheep conserved population, commercial Hu sheep population, and Chinese Merino prolific meat strain including Hu sheep blood. The results showed that six microsatellite markers were all polymorphic markers. There were 34, 33 and 29 alleles detected in three sheep populations, and the mean polymorphism information content (PIC) was 0.505 4,0.455 9 and 0.532 9, respectively. Furthermore, there were 21 alleles shared by three populations. And the genetic diversity, heterozygosity, and effective number of allele of Hu sheep from Hu Natural Source Conservative Region were higher than that of commercial population. The results showed that the genetic diversity of conservation species population was significantly higher than that of commercial population. But some rare alleles should be protected simultaneously. These findings will provide protective information of genetic diversity and Hu sheep identification.
