摘  要:为了研究凝溶胶蛋白(gelsolin,GSN)对成肌细胞分化及肌纤维形成的作用及其作用机制,试验采用蛋白导入试剂将人血浆凝溶胶蛋白导入C2C12细胞中;通过倒置显微镜定性观察细胞分化的形态学变化并应用间接免疫荧光染色法检测凝溶胶蛋白的导入情况;利用TaqMan实时定量RT-PCR方法定量分析分化前导入凝溶胶蛋白对细胞骨骼肌肌动蛋白表达水平的影响。结果表明:外源凝溶胶蛋白改变了C2C12小鼠成肌细胞的形态,使其形成发达的片状伪足或丝状伪足,分化形成的多核肌管更多;细胞分化前导入凝溶胶蛋白上调骨骼肌肌动蛋白的表达。体外试验证明C2C12小鼠成肌细胞分化前导入凝溶胶蛋白可促进肌细胞的分化及肌纤维的形成。To investigate the effects of GSN on muscle fiber formation, and initially elucidate its potential mechanism. Human plasma GSN was transduced into mouse C2C12 cells by utilizing transduction reagents; the morphologic change of celluar differentiation was observed using the inverted microscope; the transduction of GSN was assayed by indirect immunofluorescence. The expression levels of α-actin on differentiation prior to GSN transduction into C2C12 cells were quantitative detected by TaqMan real-time RT-PCR. Results were that exogenous GSN affected the morphologic of mouse C2C12 cells, and cells formed lamellipodium even filopodia,and more polynucleation myotubule. The GSN transduction before differentiation could enhance the expression levels of a-actin. These indicated that transduction GSN into C2C12 cells before differentiation can enhance myocyte differentiation.
