摘  要:目的探讨不同毒力的金黄色葡萄球菌(标准菌株ATCC25923,产肠毒素株FRS6B)感染人脐静脉内皮细胞(HUVEC)初期,细胞骨架动态变化情况。方法常规方法制备菌液,分别感染HUVEC,1、2、4h后流式细胞仪分析细胞凋亡,应用激光共聚焦荧光显微镜观察细胞骨架的动态变化。结果两种不同毒力的金黄色葡萄球菌与HUVEC共育1h后即有凋亡细胞发生,共聚焦显微镜观察可见两种细胞作用1h后细胞骨架微丝、微管均发生了改变,作用4h后细胞内聚集大量细菌,细胞骨架微丝、微管部分开始解离。结论两种金黄色葡萄球菌感染HUVEC初期,产肠毒素菌株FRS6B诱导细胞凋亡明显高于ATCC25923,细胞骨架的动态变化存在差异。Objective To investigate cell apptosis and the changes of cytoskeleton of human umbilical vein endothelial cells (HUVECs) induced by Staphylococcal aureus strains in vitro. Methods The cultured human umbilical vein HUVECs were infected by Staphylococcal aureus solutions containing standard strain ATCC2593 or enterotoxic strain FRS6B, which were prepared by conventional methods. The apoptotic cells were analyzed by flow cytometry and the changes of cytoskeleton microtubules and microfilaments were observed by confocal fluorescence microscopy 1, 2 and 4 h after treatment. Results The apoptetic HUVECs were detected by flow cytometry lh after treatment of two Staphylococcal aureus strains. The percentages of apoptotic cells were 7.90% and 6.09% after 2 and 4h treatment of ATCC25923 strain, respectively; those were 9.03% and 12.28% for FRS6B treatment. Compared with control group, the percentages of apoptotic cells were significantly higher in treatment groups. Confocal microscopy demonstrated that the cytoskeleton of infected endothelial cells started changes lh after treatment; and large amount of bacteria accumulated, skeleton microtubules and microfilaments were disaggregated 4h after treatment. Conclusion The endotoxic Staphylococcal aureus strain FRS6B can induce more apoptotic HUVECs than standard strain ATCC25923, but for induced cytoskeleton changes there are no differences between two strains.
