摘  要:目的观察实验性自身免疫性神经炎(EAN)大鼠周围神经病理、电生理和血清TNF-α及其受体P55表达变化。方法4S只SD大鼠,随机分为三组:对照组、模型1组、模型2组,EAN大鼠造模采用SP26一次性腹腔注射法诱导,分别于诱导造模成功后第14天(模型1组)及第28天(模型2组)电镜观察尾神经雪旺细胞变化,肌电图诱发电位仪检测运动神经传导速度(MCV)、感觉神经传导速度(SCV),放射免疫法检测血清TNF-α及P55的表达。结果模型1组及模型2组MCV、SCV低于对照组,模型2组MCV、SCV高于模型1组;模型1组及模型2组TNF-α、P55表达水平高于对照组,模型2组TNF-α表达水平高于模型1组;模型1组电镜下可见尾神经脱髓鞘,雪旺细胞变性坏死,模型2组神经细胞形态较模型1组部分恢复,可见凋亡雪旺细胞。结论EAN大鼠存在周围神经脱髓鞘等急性免疫反应损伤;细胞凋亡可能是EAN神经细胞损伤的主要形式;TNF-α在EAN发病中具有重要作用。Objective To investigate the pathological and electrophysiologieal alterations and TNF-α expression in peripheral nerves in rats with experimental autoimmune peripheral neuritis (EAN). Methods 45 SD rats were randomly divided into control, model 1 and model 2 groups. EAN rats were induced by linear injecting SP-26 in abdominal cavity, pathological changes were observed with electronmicroscope. Motor nerve conduction velocity (MCV) and sensory nerve conduction velocity (SCV) of coccygeal nerve were detected. Levels of TNF-α and P55 in serum were measured 14 days (model 1) and 28 days (model 2) after injection. Results Compared to control group, MCV and SCV in model 1 and model 2 were lower. However, MCV and SCV in model 2 were higher than model 1. Furthermore, compared to control group, the levels of TNF-α and P55 were higher in model land model 2. The levels of TNF-α were elevated in model 2 compared to model 1. Moreover, demyelination of coccygeal nerves and necrosis of Sehwann cells were observed in model 1. 28 days post--injection, the cells were morphologically improved in model 2 compared to model land Schwann cell appeared apoptosis. Conclusions A- cute immunologic injury may occur in peripheral nervus of rats with ENS; Apoptosis may be the main feature of peripheral nerve cell damages; TNF-α may play an important role in the pathogenesis of EAN.
