肾生方对氯化汞所致的急性肾功能衰竭大鼠模型的保护作用

摘  要:采用皮下注射HgCl2(1.5mg/kg)的方法复制急性肾功能衰竭(ARF)动物模型。选取55只大鼠随机分为正常对照组、模型组(HgCl2组)、银杏叶提取物组(GBE)和肾生方高剂量组(6.88g生药/kg)、肾生方低剂量组(3.44g生药/kg)5组,灌胃给药;测量给药前和造模后24h、48h、72h、96h、120h大鼠的24h排尿量,测定造模120h大鼠血液生化指标,测定肾脏系数和作肾组织病理学检查,探讨肾生方对HgCl2所致ARF大鼠模型的保护作用。结果显示,肾生方高剂量组能够使ARF大鼠在经历多尿期后,较快进入恢复期;能够显著降低造模后120h大鼠血清尿素(BUN)、肌酐(Cr)、钾(K+)的含量,显著减轻肾脏系数,显著减轻肾小管病变程度。肾生方能够显著减轻HgCl2所致的大鼠ARF。Acute renal failure(ARF) model was reproduced by mercuric chloride(HgCl2) subcutaneous injected in a dose of 1.5mg/kg b.w.Fifty-five wistar rats were randomly divided into 5 groups,control,model,grinkgo biloba extract(GBE),Shensheng Formula(SSF)low(3.44g/kg)and high(6.88g /kg)dose groups,and rats were administered orally.Before administration and at 24h、48h、72h、96h、120h,twenty-four urine was collected;blood biochemistry index at 120h,kidney index and kidney histopathology were detected to investigate the effects of SSF on rats with ARF induced by HgCl2.The results show that ARF rats were quickly convalescence after vsmax,SSF could reduce the concentration of blood urea nitrogen,creatinine and K+,could decrease kidney index at 120h and pathological change in kidney tubular was less severe.The results indicate that SSF could reduce injury in ARF rat induced by HgCl2.
