摘 要:目的:研究大蒜提取物二烯丙基化三硫(dially trisulfide,DATS)于体外对T24细胞侵袭转移能力的影响及其可能机制。方法:应用MTT法检测DATS对T24细胞黏附纤连蛋白能力的影响,应用Transwell小室进行人工重组基膜侵袭实验检测DATS对T24细胞侵袭能力的影响,并应用划痕愈合试验检测DATS对T24细胞迁移能力的影响。采用蛋白质印迹方法检测DATS作用后T24细胞中MMP-9蛋白的表达。结果:黏附试验表明DATS作用24h后,能有效抑制T24细胞与细胞外基质的黏附,呈剂量依赖性。划痕试验显示DATS作用不同时间对T24细胞迁移力的影响呈剂量和时间依赖性。侵袭试验表明DATS作用于T24细胞24h后,随着药物浓度的升高,T24细胞的侵袭力逐渐降低。DATS可减少T24细胞中MMP-9蛋白的表达。结论:DATS能显著抑制T24细胞的黏附、迁移和侵袭能力。DATS的侵袭转移抑制能力可能与降低MMP-9的表达有关。Objective:To examine the effects of(diallyl trisulf/de, DATS)on invasion and metasta- sis of T24 cells human bladder carcinoma and identify the possible mechanisms. Methods: For evaluating the antimetastatic activity of DATS and testing the inhibitive effect of DATS on the adhesion of T24 cells to fibronectin by MTI' assay. Wound healing vestigate the antimetastatic activities of DATS assay and the in vitro invasion assay were used to in- against T24 cells. The effect of DATS on the gene ex- pression of MMPs was examined by treating human bladder carcinoma T24 ceils with DATS. The role of MMP -9 in antimetastatic activities was analyzed by Western blotting. Results:Using the T24 human bladder cancer cell line, the DATS treatment resulted in dose - dependent inhibition of the adhesion ac-tivities was found. The cellular motility was evidently inhibited in dose - dependent and time - depend- ent manner by DATS. Similarly the results of the invasion assay in vitro also displayed that DATS was able to inhibit invasion ability of T24 cells dose - dependently. Western bloting analysis indicated that treatment of T24 cells with DATS resulted in a dose -dependent decrease in MMP -9 protein levels. Conclusion:DATS passessed a significant inhibition of cell adhesion, migration, and invasion in hu- man bladder carcinoma T24 ceils in vitro. The inhibition of invasion ability of T24 cells by DATS was attributable to decrease of the expression of MMP -9.
