摘 要:目的探讨星形胶质细胞在液压冲击损伤后蛋白谱变化。方法原代培养sD大鼠脑皮质星形胶质细胞,随机分为对照组和损伤组,损伤组给予液压冲击损伤。采用比较蛋白质组学方法,分析2者之间的差异蛋白质表达谱。结果胶质纤维酸性蛋白(GFAP)免疫组化染色鉴定,原代培养的星形胶质细胞纯度可达95%以上。液压冲击损伤使星形胶质细胞蛋白质谱表达发生了显著改变,共发现20个蛋白质点的表达发生改变,13个蛋白质得到质谱鉴定,其中表达上调的11个,分别为肌动蛋白结合蛋白、破解蛋白、磷酸甘油酸变位酶、NADH脱氢酶10亚基、膜联蛋白1、甘油醛。3.磷酸脱氢酶、热休克蛋白27ku、3-磷酸甘油酸脱氢酶、超氧化物歧化酶[Cu-Zn]、不均一核糖核蛋白、抗氧化蛋白2;表达下降的2个,分别为磷脂酰乙醇胺结合蛋白、ATP合酶D链。结论本研究发现的差异蛋白质与星形胶质细胞许多重要的细胞生理活动和功能有密切关系,包括糖代谢与能量产生、抗氧化损伤、细胞骨架重排、信号转导等,可能与损伤后星形胶质细胞发生应激有关。Objective Aim to study the characteristics ofproteomics changes in the astrocytes induced by Fluid percussion injury. Methods Primary cultures of astrocytes were prepared from cerebral hemispheres of 24 h-old rat and subsewuently divided into control group and injury group which was subjected to fluid percus- sion injury. Comparative proteomics approaches were used to study the differential protein expression profile. Results Astrocytes's proteomics changes was apparent. Dynamic proteomics changes were identified and a to- tal of 20 spots were detected in this study from the two-dimensional electrophoresis(2-DE) gels, of which 13 spots were identified via MALDI -TOF. Among them, 11 were up-regulated, namely cofilin 1, destrin, Phosphoglycerate mutase 1, NADH dehydrogenase (ubiquinone) 1 alpha subcomplex 10, annexin 1, glyceraldehydes-3-phosphate dehydrogenase, heat shock protein 27 ku, 3-phosphglycerate dehydrogenase, superoxide dismutase [Cu-Zn], het- erogeneous nuclear ribonucleoprotein A2/B1, and antioxidant protein 2. wheraeas 2 were down-regulated, namely phosphatidylethanolamine-binding protein and ATP synthase D chain. Conclusion the identified proteins in astrocytes are associated with many important cellular processes and functions such as anaerobic glycolysis ,ey- toskeleton organization, biotransformation, and signal transduction.
