摘  要:目的:探讨抗DR5单克隆抗体(mDRA-6)对小细胞肺癌NCL-H446的凋亡作用。方法:NCL-H446细胞常规培养24 h后分为对照组、抗DR5单克隆抗体(mDRA-6)及mDRA-6+顺铂处理组。以细胞形态学改变、流式细胞仪和DNA凝胶电泳分析为凋亡观察指标。结果:经mDRA-6处理的NCL-H446细胞出现核固缩、染色质边缘化和凋亡小体形成;琼脂糖凝胶电泳呈现细胞凋亡特征性DNA梯状带;细胞凋亡发生率与抗DR5单克隆抗体(mDRA-6)剂量及细胞表面DR5的表达呈相关性。结论:mDRA-6对小细胞肺癌NCL-H446的杀伤作用主要通过诱导凋亡实现的,细胞表面DR5受体表达上调可促进凋亡作用。Objective: To explore the apoptotic effect of mDRA-6 on small cell lung cancer line NCL-H446 and its mechanism.Methods: NCL-H446 cells were cultured 24 hours routinely,which were divided into the control group and the treatment group with anti-DR5 mDRA-6 and mDRA-6+ Cisplatin.The apoptosis of cells was observed by the methods of morphological changes,flow cytometry analysis and agarose gel electrophoresis of DNA.Results: mDRA-6 induced the formation of karyopycnosis,chromatic marginalization,apoptotic bodies and DNA ladder in small cell cancer line NCL-H446.The apoptotic effect was dependent on the dose and related with DR5 expressed on NCL-H446 cells.Conclusion: mDRA-6 exerts cytotoxic effect on small cell cancer line NCL-H446 by the induction of apoptosis and its apoptotic effect is promoted by up-regulated DR5 expression on NCL-H446 cell.
