摘 要:喻日成黄竹筠摘要目的:探讨不同浓度的溶血卵磷脂(LPC)对牛视网膜微血管内皮细胞转化生长因子-β2(TGF-β2)及其Ⅱ型受体表达的影响。方法:原代分离培养牛的视网膜微血管内皮细胞,加入不同浓度的LPC,分别培养6、12、24h,RT-PCR检测TGF-β2mRNA表达水平,半巢式RT-PCR检测TGF-βⅡ型受体mRNA的表达水平。结果:LPC的浓度为40μmol/L作用12h后,TGF-β2mRNA水平明显高于浓度为20μmol/L和60μmol/L。LPC浓度改变对TGF-βⅡ型受体mRNA的表达无明显影响。结论:LPC能调节牛视网膜内皮细胞TGF-β2的表达;LPC对TGF-βⅡ型受体的表达无明显影响。Objective To investigate the effects of lysophosphatidylcholine (LPC)on the expressions of transforming growth factor β2 (TGF-β2) and its receptor Ⅱ in bovine retinal endotheliial cells. Methods Bovine retinal endothelial cells were isolated and cultured in vitro with different concentrations of LPC. Levels of TGF-β2 and TGF-β receptor Ⅱ mRNA were determined by RT-PCR and heminested RT-PCR analysis in bovine retinal endothelial cells after 6,12 and 24 h. Results The level of TGF-β2 mRNA was significantly higher at 40 μmol/L than those at 20 or 60 μmol/L after 12 hours of cahivation. There was no significant effect of LPC concentrations on the expression of TGF-β3 receptor Ⅱ mRNA. Conclusion LPC can regulate the expression of TGF-β2 mRNA, not TGF-β receptor Ⅱ mRNA, in bovine retinal endothelial cells.
