摘 要:目的:研究淫羊藿总黄酮(TFE)对异丙肾上腺素所致充血性心力衰竭大鼠心肌中基质金属蛋白酶-2(MMP-2)和MMP-9表达及活性的影响。方法:取Sprague-Dawley雄性大鼠,采用皮下注射异丙肾上腺素(ISO)方法建立心力衰竭模型。将造模成功的动物随机分为模型组(n=15),美托洛尔组(n=12),TFE小剂量组(n=11),TFE中剂量组(n=10),TFE大剂量组(n=12),另取皮下注射生理盐水的正常大鼠作为正常组(n=10)。观察各组动物血流动力学变化,大鼠心肌MMP-2和MMP-9 mRNA表达水平及MMP-2和MMP-9酶活性的变化。结果:与模型组比较,淫羊藿总黄酮可逆转左心室质量指数和血流动力学参数(P<0.05或0.01),减少大鼠心肌MMP-2和MMP-9mRNA的表达并降低MMP-2和MMP-9的酶活性(P<0.05或0.01)。结论:TFE对心力衰竭大鼠心肌MMP-2和MMP-9mRNA的表达以及MMP-2和MMP-9的酶活性均有抑制作用,有助于改善心力衰竭大鼠的心肌重构。Objective To investigate the effects of total flavonoids of epimedium (TFE) on mRNA expression and activities of metalloproteinase-2(MMP-2), MMP-9 in myocardium of a rat model of congestive heart failure (CHF) induced by isoproterenol.Methods CHF was induced in male Sprague-Dawley rat model was established using subcutaneous injection of isoproterenol. Rat models of CHF were randomized into model group (n = 15), metoprolol group (n = 12), TFE-low-dose group(n = 11), TFE-medium-dose group(n = 10) and TFE-high-dose group(n = 12). Rats injected with saline were used as normal cotrols(n = 10). Left ventricular weight index (LVWI) and haemodynamic parameters of rat hears in each group were detected. MMP-2,MMP-9 mRNA expression in rat myocardium was determined by RT-PCR and real-time PCR, MMP-2, MMP-9 activities were examined by gelatin zymography assay. Results Compared with model group of CHF, TFE could dose-dependently rescure the diversities of LVWI and hemodynamic parameters(P 〈 0.05 or 〈 0.01 ). In addition, TFE could also significantly down-regulated MMP-2, MMP-9 mRNA expression (P 〈 0.05 or 0.01 ) and suppressed activities of MMP-2 and MMP-9. Conclusion TFE can suppress mRNA expression and activities of MMP-2, MMP-9 in myocardium of a rat model of CHF induced by isoproterenol.This indicates that TFE may has a protective role of preventing myocardial remodeling associated with CHF.
