探讨动脉粥样硬化斑块及龈下菌斑中检测的病原菌的同源性

摘  要:目的探讨动脉粥样硬化斑块中检测的病原菌-福赛类杆菌(T.f)、具核梭杆菌(F.n)与龈下菌斑中检测的牙周致病菌的关系。方法选择30例动脉搭桥的冠心病患者,分别取冠状动脉粥样硬化斑块及其龈下菌斑,采用牙周致病菌特异性引物,通过(polymerase chain reaction,PCR)技术,检测粥样硬化斑块及龈下菌斑中病原菌的DNA并对其扩增产物进行测序分析及比对。结果将两种细菌PCR产物的测序结果,通过blast比对,达到显著联配,它们的核苷酸相似程度为99%-100%,且符合Genbank中的标准序列。结论动脉粥样硬化斑块及龈下菌斑中检测的病原菌有同源性,说明牙周致病菌在动脉粥样硬化发生、发展过程中的可能起一定的作用。Objective To compare the periodontal microorganisms DNA(Tannerella forsythensis and Fusobacterium nucleatum) in coronary atheromatous plaques with bacterias from the subgingival plaque sample obtained from the same patient and to establish a correlation between bacteria contributing to atheromatous plaques and species associated with periodontal disease.Methods A total of 30 patients scheduled for coronary artery bypass graft were enrolled in this study.Coronary atheromatous plaques and subgingival plaque samples were examined using the polymerase chain reaction(PCR) technique by means of specific primers for periodontal bacteria.Results PCR products were sequenced and were compared with each other to ensure their alignment by blast.Conclusion The study could indicate a homology between bacteria contributing to atheromatous plaques and species associated with periodontal disease.Thus correlation may be established between coronary heart disease and periodontal disease.
