再生障碍性贫血患者的血清抑制造血活性和诱导凋亡活性及逆转对策

摘　　要：目的探讨再生障碍性贫血(再障)患者血清抑制造血活性和诱导凋亡活性及其逆转对策。方法应用甲基纤维素体外培养法、电镜、流式细胞术观察39例再障患者血清体外抑制正常人骨髓造血祖细胞增殖及促进正常人造血细胞凋亡的作用,并用不同浓度的造血细胞因子组合影响患者血清抑制造血活性和诱导凋亡活性。结果12例重型再障(SAA)和27例非重型再障(NSAA)患者血清在体外均可明显抑制正常人骨髓红系祖细胞和粒—单系祖细胞的增殖,患者血清作用后的集落细胞中有较多细胞呈现凋亡形态学改变,凋亡率明显高于对照组。SAA患者血清集落形成抑制活性和诱导凋亡活性均强于NSAA患者血清;培养体系中加入一定浓度的造血细胞因子,集落细胞产率可不同程度的恢复,集落细胞的凋亡率可不同程度的降低。结论再障患者血清具有抑制造血活性和诱导造血细胞凋亡活性,造血细胞因子可不同程度地减弱其两种活性。Objective To investigate the effect of inhibiting activity and increasing apoptosis of hematopoietic cells by sera from patients with aplastic anemia and related reversing strategy. Methods The effect of inhibiting healthy marrow hemopoietie progenitor cell proliferation of serum and promotion of hematopoietic cell apoptosis in 39 patients with aplastie anemia in vito were detected by methylcellulose in vitro culture method, electron microscope, flow cytometry. Different density of hemopoietie cytokine combination was used to influence the activity of inhibiting hemopoiesis and inducing apoptosis. Results Serum from 12 cases of severious aregenerative anemia（SAA） and 27 cases of no severious aregencrative anemia（NSAA） patients inhibited cell proliferation of nomal marrow erythro-system progenitor cell and granule-monophyly progenitor cell. There was morphology change of apoptosis in many settlement cells after the action of serum, the apopsis rate of patients was higher than that of control group. The activity of inhibiting colony forming and apoptosis-induced in SAA patients were stronger than that of NSAA. Settlement cell yield rate retrieved, apoptosis rate of settlement cell decreased in different degree, after adding hemopoietic cytokine in cultivation system. Conclusions Sera from patients with aplastic anemia can suppress the growth of normal hematopoietie progenitor cells and induce hematopoietic cells apoptosis.
