摘  要:用羧甲基化的以N,N’-甲叉双丙烯酰胺交联的烯丙基葡聚糖(简称CM-CADB)凝胶酶载体用树脂为新型载体,将谷氨酸脱羧酶固定在CM-CADB树脂上。研究了固定化谷氨酸脱羧酶和自由酶与pH、温度的依存性,使用热稳定性和寿命。经测定谷氨酸脱羧酶经过固定化后,最适pH向碱性方向偏移02个单位,pH为52,最适温度为48℃。固定化酶使用热稳定性比自由酶下降了5℃,但固定化酶的使用寿命得到显著改善,经测定固定化谷氨酸脱羧酶在4℃保存时,半衰期为127天。The immobilized L-glutamic decarboxylase on the CM-CADB (carboxymethyl-copolymer of allyl dextran and N,N'-methylene-bisacrylamide) ion exchange resin has been studied in this paper. The dependence of the activity of immobilized GDC and free enzyme on the pH, temperature was studied. The thermostability and the longevity were investigated. It is observed that the optimum pH(5 2) is 0 2 more than that of the free GDC(5 0). The optimum temperature is found to be around 48℃. The critical temperature of free GDC and immobilized GDC is respectively around 50℃and 45℃. The operational stability of immobilized GDC was examined. The half-life of the activity is 127 days, when the immobilized enzyme was kept in 4℃.
